Abstract: Distortion of Left Ventricle (LV) external anatomy is related to some dysfunctions, such as hypertrophy. The architecture of myocardial fibers determines LV electromechanical activation patterns as well as mechanics. Thus, their joined modelling would allow the design of specific interventions (such as peacemaker implantation and LV remodelling) and therapies (such as resynchronization). On one hand, accurate modelling of external anatomy requires either a dense sampling or a continuous infinite dimensional approach, which requires non-Euclidean statistics. On the other hand, computation of fiber models requires statistics on Riemannian spaces. Most approaches compute separate statistical models for external anatomy and fibers architecture. In this work we propose a general mathematical framework based on differential geometry concepts for computing a statistical model including, both, external and fiber anatomy. Our framework provides a continuous approach to external anatomy supporting standard statistics. We also provide a straightforward formula for the computation of the Riemannian fiber statistics. We have applied our methodology to the computation of complete anatomical atlas of canine hearts from diffusion tensor studies. The orientation of fibers over the average external geometry agrees with the segmental description of orientations reported in the literature.

In G Protein-Coupled Receptors – Modeling and Simulation Advances in Experimental Medicine and Biology (Vol. 796, pp. 159–181). Springer Netherlands.
Abstract: Empirical and mechanistic models differ in their approaches to the analysis of pharmacological effect. Whereas the parameters of the former are not physical constants those of the latter embody the nature, often complex, of biology. Empirical models are exclusively used for curve fitting, merely to characterize the shape of the E/[A] curves. Mechanistic models, on the contrary, enable the examination of mechanistic hypotheses by parameter simulation. Regretfully, the many parameters that mechanistic models may include can represent a great difficulty for curve fitting, representing, thus, a challenge for computational method development. In the present study some empirical and mechanistic models are shown and the connections, which may appear in a number of cases between them, are analyzed from the curves they yield. It may be concluded that systematic and careful curve shape analysis can be extremely useful for the understanding of receptor function, ligand classification and drug discovery, thus providing a common language for the communication between pharmacologists and medicinal chemists.

Abstract: Two-state and operational models of both agonism and allosterism are compared to identify and characterize common pharmacological parameters. To account for the receptor-dependent basal response, constitutive receptor activity is considered in the operational models. By arranging two-state models as the fraction of active receptors and operational models as the fractional response relative to the maximum effect of the system, a one-by-one correspondence between parameters is found. The comparative analysis allows a better understanding of complex allosteric interactions. In particular, the inclusion of constitutive receptor activity in the operational model of allosterism allows the characterization of modulators able to lower the basal response of the system; that is, allosteric modulators with negative intrinsic efficacy. Theoretical simulations and overall goodness of fit of the models to simulated data suggest that it is feasible to apply the models to experimental data and constitute one step forward in receptor theory formalism.

Abstract: The study of mental workload becomes essential for human work efficiency, health conditions and to avoid accidents, since workload compromises both performance and awareness. Although workload has been widely studied using several physiological measures, minimising the sensor network as much as possible remains both a challenge and a requirement.
Electroencephalogram (EEG) signals have shown a high correlation to specific cognitive and mental states like workload. However, there is not enough evidence in the literature to validate how well models generalize in case of new subjects performing tasks of a workload similar to the ones included during model’s training.
In this paper we propose a binary neural network to classify EEG features across different mental workloads. Two workloads, low and medium, are induced using two variants of the N-Back Test. The proposed model was validated in a dataset collected from 16 subjects and shown a high level of generalization capability: model reported an average recall of 81.81% in a leave-one-out subject evaluation.

In Association for Computing Machinery (Ed.), 14th International Symposium on Applied Sciences in Biomedical and Communication Technologies. Barcelona, Spain.
Abstract: Deep understanding of myocardial structure of the heart would unravel crucial knowledge for clinical and medical procedures. The MIOCARDIA project is a multidisciplinary project in cooperation with l'Hospital de la Santa Creu i de Sant Pau, Clinica la Creu Blanca and Barcelona Supercomputing Center. The ultimate goal of this project is defining a computational model of the myocardium. The model takes into account the deep interrelation between the anatomy and the mechanics of the heart. The paper explains the workflow of the MIOCARDIA project. It also introduces a multiresolution reconstruction technique based on DT-MRI streamlining for simplified global myocardial model generation. Our reconstructions can restore the most complex myocardial structures and provides evidences of a global helical organization.

Misael Rosales, Petia Radeva, Oriol Rodriguez-Leon, & Debora Gil. (2009). Modelling of image-catheter motion for 3-D IVUS. MIA - Medical image analysis, 13(1), 91–104.
Abstract: Three-dimensional intravascular ultrasound (IVUS) allows to visualize and obtain volumetric measurements of coronary lesions through an exploration of the cross sections and longitudinal views of arteries. However, the visualization and subsequent morpho-geometric measurements in IVUS longitudinal cuts are subject to distortion caused by periodic image/vessel motion around the IVUS catheter. Usually, to overcome the image motion artifact ECG-gating and image-gated approaches are proposed, leading to slowing the pullback acquisition or disregarding part of IVUS data. In this paper, we argue that the image motion is due to 3-D vessel geometry as well as cardiac dynamics, and propose a dynamic model based on the tracking of an elliptical vessel approximation to recover the rigid transformation and align IVUS images without loosing any IVUS data. We report an extensive validation with synthetic simulated data and in vivo IVUS sequences of 30 patients achieving an average reduction of the image artifact of 97% in synthetic data and 79% in real-data. Our study shows that IVUS alignment improves longitudinal analysis of the IVUS data and is a necessary step towards accurate reconstruction and volumetric measurements of 3-D IVUS.

Abstract: Even using objective measures from DT-MRI no consensus about myocardial architecture has been achieved so far. Streamlining provides good reconstructions at low level of detail, but falls short to give global abstract interpretations. In this paper, we present a multi-resolution methodology that is able to produce simplified representations of cardiac architecture. Our approach produces a reduced set of tracts that are representative of the main geometric features of myocardial anatomical structure. Experiments show that fiber geometry is preserved along reductions, which validates the simplified model for interpretation of cardiac architecture.

Abstract: The operational model of agonism assumes that the maximum effect a particular receptor system can achieve (the Em parameter) is fixed. Em estimates are above but close to the asymptotic maximum effects of endogenous agonists. The concept of Em is contradicted by superagonists and those positive allosteric modulators that significantly increase the maximum effect of endogenous agonists. An extension of the operational model is proposed that assumes that the Em parameter does not necessarily have a single value for a receptor system but has multiple values associated to multiple active receptor conformations. The model provides a mechanistic link between active receptor conformation and agonist efficacy, which can be useful for the analysis of agonist response under different receptor scenarios.

Abstract: Deep understanding of myocardial structure of the heart would unravel crucial knowledge for clinical and medical procedures. Although the muscular architecture of the heart has been debated by countless researchers, the controversy is still alive. Diffusion Tensor MRI, DT-MRI, is a unique imaging technique for computational validation of the muscular structure of the heart. By the complex arrangement of myocites, existing techniques can not provide comprehensive descriptions of the global muscular architecture. In this paper we introduce a multiresolution reconstruction technique based on DT-MRI streamlining for simplified global myocardial model generation. Our reconstructions can restore the most complex myocardial structures and indicate a global helical organization

Debora Gil, Oriol Rodriguez-Leon, Petia Radeva, & Josepa Mauri. (2008). Myocardial Perfusion Characterization From Contrast Angiography Spectral Distribution. IEEE Transactions on Medical Imaging, 27(5), 641–649.
Abstract: Despite recovering a normal coronary flow after acute myocardial infarction, percutaneous coronary intervention does not guarantee a proper perfusion (irrigation) of the infarcted area. This damage in microcirculation integrity may detrimentally affect the patient survival. Visual assessment of the myocardium opacification in contrast angiography serves to define a subjective score of the microcirculation integrity myocardial blush analysis (MBA). Although MBA correlates with patient prognosis its visual assessment is a very difficult task that requires of a highly expertise training in order to achieve a good intraobserver and interobserver agreement. In this paper, we provide objective descriptors of the myocardium staining pattern by analyzing the spectrum of the image local statistics. The descriptors proposed discriminate among the different phenomena observed in the angiographic sequence and allow defining an objective score of the myocardial perfusion.
