Abstract |
Individuals diagnosed with Mild Cognitive Impairment (MCI) have shown an increased risk of developing Alzheimer’s Disease (AD). As such, early identification of dementia represents a key prognostic element, though hampered by complex disease patterns. Increasing efforts have focused on Machine Learning (ML) to build accurate classification models relying on a multitude of clinical/imaging variables. However, ML itself does not provide sensible explanations related to the model mechanism and feature contribution. Explainable Artificial Intelligence (XAI) represents the enabling technology in this framework, allowing to understand ML outcomes and derive human-understandable explanations. In this study, we aimed at exploring ML combined with MRI-based features and XAI to solve this classification problem and interpret the outcome. In particular, we propose a new method to assess the robustness of feature rankings provided by XAI methods, especially when multicollinearity exists. Our findings indicate that our method was able to disentangle the list of the informative features underlying dementia, with important implications for aiding personalized monitoring plans. |

Abstract |
Recent breakthroughs in Artificial Intelligence, Deep Learning and Document Image Analysis and Recognition have significantly eased the creation of digital libraries and the transcription of historical documents. However, for documents in rare scripts with few labelled training data available, current Handwritten Text Recognition (HTR) systems are too constraint. Moreover, research on HTR often focuses on technical aspects only, and rarely puts emphasis on implementing software tools for scholars in Humanities. In this article, we describe, compare and analyse different transcription methods for rare scripts. We evaluate their performance in a real use case of a medieval manuscript written in the runic script (Codex Runicus) and discuss advantages and disadvantages of each method from the user perspective. From this exhaustive analysis and comparison with a fully manual transcription, we raise conclusions and provide recommendations to scholars interested in using automatic transcription tools. |

Abstract |
Low resource Handwritten Text Recognition (HTR) is a hard problem due to the scarce annotated data and the very limited linguistic information (dictionaries and language models). This appears, for example, in the case of historical ciphered manuscripts, which are usually written with invented alphabets to hide the content. Thus, in this paper we address this problem through a data generation technique based on Bayesian Program Learning (BPL). Contrary to traditional generation approaches, which require a huge amount of annotated images, our method is able to generate human-like handwriting using only one sample of each symbol from the desired alphabet. After generating symbols, we create synthetic lines to train state-of-the-art HTR architectures in a segmentation free fashion. Quantitative and qualitative analyses were carried out and confirm the effectiveness of the proposed method, achieving competitive results compared to the usage of real annotated data. |

Abstract |
Human multi-limb segmentation in RGB images has attracted a lot of interest in the research community because of the huge amount of possible applications in fields like Human-Computer Interaction, Surveillance, eHealth, or Gaming. Nevertheless, human multi-limb segmentation is a very hard task because of the changes in appearance produced by different points of view, clothing, lighting conditions, occlusions, and number of articulations of the human body. Furthermore, this huge pose variability makes the availability of large annotated datasets difficult. In this paper, we introduce the HuPBA8k+ dataset. The dataset contains more than 8000 labeled frames at pixel precision, including more than 120000 manually labeled samples of 14 different limbs. For completeness, the dataset is also labeled at frame-level with action annotations drawn from an 11 action dictionary which includes both single person actions and person-person interactive actions. Furthermore, we also propose a two-stage approach for the segmentation of human limbs. In a first stage, human limbs are trained using cascades of classifiers to be split in a tree-structure way, which is included in an Error-Correcting Output Codes (ECOC) framework to define a body-like probability map. This map is used to obtain a binary mask of the subject by means of GMM color modelling and GraphCuts theory. In a second stage, we embed a similar tree-structure in an ECOC framework to build a more accurate set of limb-like probability maps within the segmented user mask, that are fed to a multi-label GraphCut procedure to obtain final multi-limb segmentation. The methodology is tested on the novel HuPBA8k+ dataset, showing performance improvements in comparison to state-of-the-art approaches. In addition, a baseline of standard action recognition methods for the 11 actions categories of the novel dataset is also provided. |

Abstract |
We present in this paper a novel lumen centre detection for gastrointestinal and respiratory endoscopic images. The proposed method is based on the appearance and geometry of the lumen, which we defined as the darkest image region which centre is a hub of image gradients. Experimental results validated on the first public annotated gastro-respiratory database prove the reliability of the method for a wide range of images (with precision over 95 %). |

In somatic cells, chromosomes occupy specific nuclear regions called chromosome territories which are involved in the
maintenance and regulation of the genome. Preliminary data in male germ cells also suggest the importance of chromosome
territoriality in cell functionality. Nevertheless, the specific characteristics of testicular tissue (presence of different
cell types with different morphological characteristics, in different stages of development and with different ploidy)
makes difficult to achieve conclusive results. In this study we have developed a methodology to approach the threedimensional
study of all chromosome territories in male germ cells from C57BL/6J mice (Mus musculus). The method
includes the following steps: i) Optimized cell fixation to obtain an optimal preservation of the three-dimensionality cell
morphology, ii) Chromosome identification by FISH (Chromoprobe Multiprobe® OctoChrome™ Murine System; Cytocell)
and confocal microscopy (TCS-SP5, Leica Microsystems), iii) Cell type identification by immunofluorescence
iv) Image analysis using Matlab scripts, v) Numerical data extraction related to chromosome features, chromosome
radial position and chromosome relative position. This methodology allows the unequivocally identification and the
analysis of the chromosome territories of all spermatogenic stages. Results will provide information about the features
that determine chromosomal position, preferred associations between chromosomes, and the relationship between chromosome
positioning and genome regulation. |

Abstract |
Computer-aided diagnosis (CAD) is a tool with great potential to help endoscopists in the tasks of detecting and histologically classifying colorectal polyps. In recent years, different technologies have been described and their potential utility has been increasingly evidenced, which has generated great expectations among scientific societies. However, most of these works are retrospective and use images of different quality and characteristics which are analysed off line. This review aims to familiarise gastroenterologists with computational methods and the particularities of endoscopic imaging, which have an impact on image processing analysis. Finally, the publicly available image databases, needed to compare and confirm the results obtained with different methods, are presented. |

Chromosome territoriality is not random along the cell cycle and it is mainly governed by intrinsic chromosome factors and gene expression patterns. Conversely, very few studies have explored the factors that determine chromosome territoriality and its influencing factors during meiosis. In this study, we analysed chromosome positioning in murine spermatogenic cells using three-dimensionally fluorescence in situ hybridization-based methodology, which allows the analysis of the entire karyotype. The main objective of the study was to decipher chromosome positioning in a radial axis (all analysed germ-cell nuclei) and longitudinal axis (only spermatozoa) and to identify the chromosomal factors that regulate such an arrangement. Results demonstrated that the radial positioning of chromosomes during spermatogenesis was cell-type specific and influenced by chromosomal factors associated to gene activity. Chromosomes with specific features that enhance transcription (high GC content, high gene density and high numbers of predicted expressed genes) were preferentially observed in the inner part of the nucleus in virtually all cell types. Moreover, the position of the sex chromosomes was influenced by their transcriptional status, from the periphery of the nucleus when its activity was repressed (pachytene) to a more internal position when it is partially activated (spermatid). At pachytene, chromosome positioning was also influenced by chromosome size due to the bouquet formation. Longitudinal chromosome positioning in the sperm nucleus was not random either, suggesting the importance of ordered longitudinal positioning for the release and activation of the paternal genome after fertilisation. |

Abstract |
In most eukaryotes, pairing of homologous chromosomes is an essential feature of meiosis that ensures homologous recombination and segregation. However, when the pairing process begins, it is still under investigation. Contrasting data exists in Mus musculus, since both leptotene DSB-dependent and preleptotene DSB-independent mechanisms have been described. To unravel this contention, we examined homologous pairing in pre-meiotic and meiotic Mus musculus cells using a threedimensional fuorescence in situ hybridization-based protocol, which enables the analysis of the entire karyotype using DNA painting probes. Our data establishes in an unambiguously manner that 73.83% of homologous chromosomes are already paired at premeiotic stages (spermatogonia-early preleptotene spermatocytes). The percentage of paired homologous chromosomes increases to 84.60% at mid-preleptotene-zygotene stage, reaching 100% at pachytene stage. Importantly, our results demonstrate a high percentage of homologous pairing observed before the onset of meiosis; this pairing does not occur randomly, as the percentage was higher than that observed in somatic cells (19.47%) and between nonhomologous chromosomes (41.1%). Finally, we have also observed that premeiotic homologous pairing is asynchronous and independent of the chromosome size, GC content, or presence of NOR regions. |
